This tissue cell lysis reagent uses a 25 mM N-diglycine, 150 mM sodium chloride solution (pH 7.6) containing a proprietary detergent to maximize the passage of Efficiency of homogenization in solubilizing proteins from mammalian tissue samples. This simple component is compatible with a wide range of additives (e.g., various protease inhibitors, salts, reducing agents, chelating agents), providing versatility for many different sample types and lysis applications. Cell lysates prepared using T-PER reagent can be directly combined with various reporter gene assays (e.g., luciferase, β-galactosidase, chloramphenicol acetyltransferase), protein kinase assays (e.g., PKA, PKC, tyrosine acid kinase), immunoassays (e.g., Western blot, ELISA, RIA) and/or protein purification procedures.
Efficient homogeneous extraction of tissue proteins from different samples is challenging because of differences in tissue composition and extraction methods. Fibrous and collagenous tissues (such as heart and kidney) make protein extraction by freeze-thaw or mortar and pestle methods inefficient without the use of detergents. In contrast, soft tissues such as the spleen and liver require milder extraction protocols to achieve better results. T-PER reagents are designed for use with all tissues, regardless of mechanical extraction method. The mild detergent solubilizes proteins without affecting functionality, and the reagent's neutral pH and salt concentration allow for more efficient extraction from cellular compartments, producing homogeneous lysates.
The protein yield of T-PER reagent in fresh and frozen tissues was verified. Frozen tissue and freeze-thaw extraction protocols can predictably achieve higher protein yields. In addition, T-PER reagent has been shown to efficiently extract from multiple cellular compartments such as plasma membrane (Na-K+ ATPase, pan-cadherin), cytosol (HSP90, MAPK), mitochondria (COXIV), and nucleus (HDAC1) Target protein as a tissue-specific marker in brain (synaptophysin) and heart tissue (troponin T cardiac isoform).
