The blocked magnetic bead surface is derivatized using nitrilotriacetic acid (NTA) chelating groups with di- Valent nickel ions (Ni2+). Immobilized Metal Affinity Chromatography (IMAC) beads provide high binding capacity and extremely low background. HisPur Ni-NTA magnetic beads can be used with a magnetic stand for manual operation, or can be used on automated platforms, such as Thermo Scientific KingFisher instruments, to meet high-throughput needs.

HisPur Ni-NTA is used for small-scale affinity purification and high-throughput screening of recombinant His-tagged proteins. Polyhistidine tags are extremely commonly used affinity tags, usually consisting of 6 consecutive histidine residues (6xHis). These tagged proteins are overexpressed in many different systems (most commonly bacteria) and purified from cell lysates such as those prepared using B-PER Bacterial Protein Extraction Reagent. His-tagged proteins are purified using NTA chelates with nickel ions that match histidine side chains. NTA chelates contain four metal binding sites, providing low metal ion leaching rates and high binding capacity. The protocol for HisPur Ni-NTA magnetic beads has been optimized to provide high purity isolation of His-tagged proteins. Performance is comparable to or higher than Ni-NTA magnetic beads provided by other suppliers.
