Whether chemiluminescent Western blotting or DNA or RNA autoradiography, the Thermo Scientific Pierce Background Eliminator Kit can be used during development Quickly and easily remove blurred areas and artifacts from X-ray film to correct overexposure of prints.

Background Eliminator Kit Features:

For overexposed Removal of silver emulsion
Recoverable and Preservable Data—Reduces the resulting signal uniformly across the entire film without changing the relative intensity (optical density) of the bands
Optimization Signal-to-Noise Ratio—Allows over-developed film to be re-adjusted to resolve background blur areas, spots, and oversaturated bands for the best possible image
Saving Time and Resources—No need Repeat the experiment or re-expose with new film to get the best image

This two-reagent kit is easy to use, as shown in the video below. Mix the two reagents (1:1 ratio) and immerse a piece of developed film (recent or old) into the working solution. Observe for 1 to 5 minutes until the desired image appears, then stop the reaction by rinsing the film in water. Pierce Film Background Eliminator inverts the developed signal evenly across the entire film and preserves relative optical density values, allowing analysis and publication of results to remain valid and feasible.

Applications:
• Develop X-ray film by any type of enhanced chemiluminescence Western blot, whether Thermo Scientific SuperSignal Pierce ECL or other commercial substrate
• Develop X-ray films from any chemiluminescence or radioactive gel shift assay, including the Thermo Scientific LightShift Chemiluminescence EMSA Kit for DNA or RNA detection
• Develop from chemiluminescence or isotope (32P or 35S) Northern or Southern blotting Develop autoradiographic film with any method, including the Thermo Scientific North2South Chemiluminescence Detection Kit
• Any developed silver emulsion film (whether blue, clear, single or double sided), regardless of exposure method
< br>High background, shadows, overexposed bands and spots are problems inherent to film exposure. Higher background and shadowing can be caused by overexposure, improper use of blocking buffer, or inappropriate enzyme-labeled probe or antibody concentrations. Overexposed bands often occur when the concentration of enzyme-labeled probes or antibodies used is too high or the film is exposed for too long. Spots and shadows are produced when the enzyme conjugate forms complexes and precipitates on the blot. Pierce Background Eliminator corrects all of these problems without re-exposing the blot to film or redoing the entire experiment. Working solution can be used with newly exposed film or film that has been stored for several years. Additionally, this product can be used with any brand of film.
