Cell Proliferation Dye eFluor™ 450 is a purple fluorescent dye that can be used to monitor single cell division. This fluorescent dye binds to any cellular protein containing a primary amine, and as cells divide, the dye is evenly distributed among daughter cells, as measured by the sequential halving of the dye's fluorescence intensity. Up to 7 passages of cells were observed. Cells labeled with Cell Proliferation Dye eFluor™ 450 are fixed and permeabilized for intracellular target analysis using standard formaldehyde-containing fixatives and saponin-based permeabilization buffers such as the Foxp3 Transcription Factor Staining Buffer Kit (Cat. No. 00-5523) or IC Fixation Buffer (Cat. No. 00-8222) and Permeabilization Buffer (10X) (Cat. No. 00-8333).
Cell Proliferation Dye eFluor™ 450 has an excitation peak of 409 nm and can be detected using a 450/50 bandpass filter (equivalent to eFluor™ 450 or Pacific Blue™), making it compatible with GFP-based App compatible.
Cell Proliferation Dye eFluor™ 450 is available as a lyophilized powder. Each vial of lyophilized powder can be reconstituted with 165 µL of anhydrous DMSO to a stock solution with a concentration of 10 mM; after reconstitution, it should be used within 6 months and stored together with a desiccant in the dark at -20 at °C. Avoid freezing and thawing.
