The pHrodo iFL Red Micro Protein Labeling Kit and the pHrodo iFL Green Micro Protein Labeling Kit provide the reagents needed to easily label 3 micro-volume purified protein or antibody samples (20–100 µg) using pHrodo iFL Red or pHrodo iFL Green dyes. These kits contain amine-reactive, pH-sensitive pHrodo iFL Red or Green STP ester dyes that covalently bind to free lysine anywhere in antibodies and proteins.

pHrodo iFL Red and pHrodo iFL Green dyes are improved versions of pHrodo Red and pHrodo Green dyes, optimized to produce bright and soluble bioconjugates suitable for antibody internalization, endocytosis and phagocytosis role studies. They are also more soluble than the original pHrodo Red and pHrodo Green dyes, making them useful for labeling antibodies that precipitate from solution during conjugation. pHrodo iFL Red and pHrodo iFL Green dyes are non-fluorescent outside cells, but will fluoresce strongly in red or green as the pH of their surrounding environment becomes more acidic (such as in phagosomes and endosomes).

The pHrodo Deep Red Mammalian and Bacterial Cell Labeling Kit provides the reagents needed to easily label living cells (mammalian or bacterial) to study phagocytosis and endocytosis. It is optimized for distinguishing phagocytic and non-phagocytic cells because the pH-sensitive, low-background pHrodo Deep Red dye has no signal under neutral conditions and fluoresces only when exposed to an acidic environment. This unique property of pHrodo Deep Red enables rapid assay development and increases the certainty of results in studies of antibody endocytosis and phagocytosis pathways. pHrodo Deep Red dye helps better distinguish internalized substances outside the cell because it has a pKa of approximately 5 and does not fluoresce until it enters late endosomes and lysosomes. pHrodo Deep Red dye is detectable using the Cy5 fluorescence filter assembly and is validated for use in a variety of applications including flow cytometry, fluorescence microscopy and high content analysis (HCA).
