Thermo Scientific X-Gal (5-bromo-4-chloro-3-indolyl-β-D-galacto Pyranoside) is an inert chromogenic substrate for β-galactosidase, which hydrolyzes X-Gal to produce colorless galactose and 4-chloro-3-bromo-indigo, forming a dark blue precipitate. IPTG induces the lacZ gene, leading to hydrolysis of X-Gal and formation of blue colonies (see Supporting Data).

X-Gal solution (ready to use) is a stable, 0.22 μm membrane filtered solution that can be used directly with IPTG for blue-white colony screening.
